摘 要:
目的 传统的方法分离培养军团菌,时间长,价格贵,培养较困难,本文建立多重PCR方法,快速检测空调冷却塔水中军团菌属和嗜肺军团菌种。方法 利用军团菌属保守基因片段16SrRNA和嗜肺军团型菌巨噬细胞感染增强因子(mip)基因序列的特异性引物,对空调冷却塔水样中分离的疑似军团菌株进行特异性扩增,并且为了增强敏感性,运用递减温度PCR扩增。结果:PCR扩增的阳性结果与生化培养和血清学鉴定为军团菌的菌株结果—致。结论:与传统的分离培养方法相比,多重PCR方法检测军团菌,快速敏感,成本低,具有较好的特异性。[著者文摘]

Abstract:
Objective It is long time and expensive to identify Legionella pneumophila with classic cutivating method .so it is difficult to incubate Legionella pneumophila. To explore the contamination by Legionella pneumophila in coolingwater of air conditioning system rapidly and conveniently, we developed the multiplex-polymerase chain reaction method. Method With primers designed according to,the sequences of 16SrRNA gene and mip gene of Legionella pneumophila we amplify 16SrRNA gene fragment and mip sequence, so detected suspicions Legionella pneumophila in water samples.What is more, we reduce temperature gradually method to incease sensitivity of detection. Result All the tested Legionella pneumophila strains with biochemical and serological method had positive PCR result . Conclusion Multiplex-PCR method of identification Legionella pneumophila shows high specificity and sensitivity and meanwhile can be completed in a relative short time surely comparing with classic incubating method, this method can be largely available and has a potential value in identifying Legionella pneumophila from water sample.[著者文摘]
