海水淹溺兔肺泡巨噬细胞TNF-α mRNA的动态表达及地塞米松干预作用

摘  要:

目的:观察海水淹溺后家兔肺泡巨噬细胞内TNF-α mRNA的动态表达规律,并探讨地塞米松干预对TNF-α mRNA表达的影响。方法:采用气管切开插入塑料导管、向气管内灌海水4ml/kg,双肺自主通气的方法模拟海水淹溺造成急性肺损伤模型。于淹溺后0min、30min、60min、90min、120min五个观察时间点进行血气分析,并采集肺组织,逆转录一聚合酶链反应(RT-PCR)检测肺泡巨噬细胞中TNF-α mRNA的表达。结果:海水淹溺兔急性肺损伤各时相点肺组织中性粒细胞大量浸润,淹溺后TNF-α mRNA表达显著增高,90分钟后达最高峰,而地塞米松可使之降低(P〈0.01)。结论:海水淹溺急性肺损伤可能与肺泡巨噬细胞内TNF-α mRNA高表达有关。地塞米松可降低TNF-α mRNA的高表达,减轻肺损伤程度。[著者文摘]

Abstract:

Objective: To observe the dynamic change in the expression of TNF-α mRNA expression in pulmonary alveolar macrophage (AM) of in rabbits drowned by with sea water drowning, and explore the effects of dexamethasone injection on the expression of TNF-α mRNA. Methods: The drowning model was established by pouring sea water (4ml/kg) to the trachea of rabbits using plastic tube during spontaneous breathing of bilateral lung auto ventilated, to simulate the process of drowning. The expression of TNF-α mRNA in pulmonary alveolar macrophages of rabbits was examined by reverse-transcription polymerase chain reaction (RT-PCR) after being drowned 0min, 30min, 60 rain, 90 min and 120min respectively. Lung tissues were examined by histopathology, and the blood gas analysis was determinated. Results: ARer drowning, the infiltration of PMN and the expression of TNF-α mRNA increased significantly, and reached highest after 90 minutes drowning. Dexamethasone inhibitsthe expression of TNF-α mRNA and extenuates acute lung injury. Conclusion: For sea water respiratory distress syndrome (SW-RDS), acute lung damage at early stage might be related to the high expression of TNF-α mRNA in AM. Dexamethasone can significantly decrease the expression of TNF-α mRNA in the alveolar macrophage (P〈0.01 ), and extenuate acute lung injury.[著者文摘]
