人骨形态蛋白-2对椎间盘细胞蛋白多糖和Ⅱ型胶原的影响

摘  要:

目的 研究人骨形态蛋白(human bone morphogenetic protein-2,hBMP-2)对体外培养人腰椎间盘髓核细胞Ⅱ型胶原和aggrecan的影响。方法 人退变髓核细胞体外分离培养,通过免疫组织化学鉴定椎间盘细胞。利用ELISA法检测对照组和不同剂量hBMP-2(10ng/ml和100ng/ml)组Ⅱ型胶原和aggrecan的表达水平。用RT-PCR检测细胞Ⅱ型胶原和aggrecan的mRNA表达水平。结果 加入hBMP-2后Ⅱ型胶原和aggrecan表达增加,并存在剂量依赖关系(P〈0.01)。在第6天RT-PCR结果显示hBMP-2组aggrecan和Ⅱ型胶原mRNA的表达高于对照组,并且随剂量增高aggrecan和Ⅱ型胶原mRNA的表达量逐渐增加。结论 hBMP-2可促进体外培养人腰椎间盘细胞合成代谢,Ⅱ型胶原和aggrecan表达量增加,并存在剂量依赖关系,提示hBMP-2可能对退变椎间盘具有修复功能。[著者文摘]

EFFECT OF HUMAN BONE MORPHOGENETIC PROTEIN-2 ON AGGRECAN AND COLLAGEN TYPE Ⅱ IN LUMBAR INTERVERTEBRAL DISC CELLS

Abstract:

Objective To study the effects of human bone morphogenetic protein 2 (hBMP-2) on aggrecan and collagen type Ⅱ in lumbar intervertebral disc cells. Methods Human intervertebral disc cells were isolated from the.nucleus pulpous of lumbar discs. The cells were cultured in vitro and identified by immunohistochemical staining. The cells were grown in monolayer and treated with hBMP-2 (10ng/ml and 100ng/ml) in the Dulbecco's Modified Eagle Medium and Ham's F-12 Medium in vitro. On the third and sixth day after h:BMP-2 treatment, the cell supernatant was collected. The expression of aggrecan and collagen type Ⅱ Was determined by enzyme-linked immunosorbent assay (ELISA). On the sixth day after hBMP-2 treatment, mRNA was extracted for reverse transcription polymerase chain reaction (RT- PCR), and the differences between the control group and the culture groups treated with hBMP-2 (10ng/ ml or 100ng/ml) was detected in the expression of aggrecan and collagen type Ⅱ mRNA. Results The expression of aggrecan and collagen type Ⅱ in these cells was gradually increased in a dose dependent manner after treatment with different doses of hBMP-2. Conclusion The results of this study show that hBMP-2 is efficient to accelerates proteoglycan and collagen type Ⅱ synthesis, so hBMP-2 may play an important role in the repair process of disc degeneration.[著者文摘]
