猪GV期与MⅡ期卵母细胞的超微结构特征

摘 要:
采用扫描电镜（SEM）和透射电镜（TEM）观察并比较猪未成熟（GV期）与体外成熟（MⅡ期）卵母细胞的超微结构特征。结果发现：①猪GV期卵母细胞外紧密包被着卵丘细胞，卵丘细胞突起伸人透明带内，透明带呈不规则蜂窝状；卵母细胞微绒毛相对较长、数量较多，并穿人透明带内；卵母细胞呈充满状态，卵周隙不明显；胞质内高尔基复合体发达，线粒体成簇分布于脂滴附近；脂滴表现为均质和不均质两种形式，大多以均质脂滴形式存在，脂滴周围有不连续的滑面内质网。②MⅡ期卵母细胞周围，卵丘细胞结构松散、扩展；卵母细胞微绒毛数量减少、变短、变粗，并从透明带内撤回；透明带上峰窝状小孔增多，小梁突起更为明显，卵周隙明显；线粒体仍成簇伴随脂滴分布，脂滴以不均质型为主，呈明显的絮状结构，脂滴外有不连续的滑面内质网；皮质颗粒以单层排列于质膜下。猪卵母细胞成熟前后会出现一些规律性变化，具有不同的超微结构特征。[著者文摘]

Ultra-structure of Porcine GV-and/VIII-oocytes

Abstract:
The ultra-structure of both porcine immature （ GV stage） and mature in vitro （ Metaphase Ⅱ , MⅡ ） oocytes were observed by scanning electron microscopy （ SEM ） and transmission electron microscopy （ TEM ）. The cumulus ceils enclosed each GV-oocyte compactly and their protuberances stretched into zona pellucida of the oocyte. The zona pellucida appeared a mesh-like structure with numerous fenestrations together with an uneven surface with a microtrabecular appearance. Long and abundant microvilli on the oolemma surface extended into the zona pellucida. The perivitelline space between GV-oocytes was not discernable. Inside the cytoplasm of GV-oocytes ample Golgi complexes and the mitochondria with a few clear ridges distributed next to lipid drops in clusters were observeg. Homogeneous droplets dominated the lipid droplets which were surrounded by smooth endoplasmic reticulum. All around MⅡ -oocytes ,the cumulus ceils became expended. The microvilli looked tubby and withdrew from zona pellucida. The microtrabecular on the surface of zona pellucida and the perivitelline space between MⅡ-oocytes could be observed distinctly. The inhomogeneous lipid droplets presented dominantly in global shape with more floccules. Clusters of mitochondria lied near the lipid droplets . The cortical granules were arranged beneath the oolemma membrane. The results indicated that before and after maturation, porcine oocytes did behave some regular alterations in ultra-structure.[著者文摘]
