摘 要:
目的:探讨神经干细胞(NSC)在分化过程中生长相关蛋白43(GAP-43)基因的表达及其意义。方法:NSC提取于新生鼠的海马区,经培养及Nestin免疫细胞化学鉴定,用RT-PCR和免疫组化方法观察GAP-43基因在NSC分化过程中不同时间点的表达。结果:GAP-43mRNA在NSC处于克隆球阶段未检测到其表达,开始分化12h可检测到其低表达,并逐渐升高,3d表达量达到高峰,5d表达量下降。GAP-43最初在NSC分化1d的胞质内表达并逐渐升高,5d表达增强,于细胞的突起中明显,7d时GAP-43反应减弱并逐渐消失。结论:在神经干细胞分化为神经元过程中GAP-43基因参与引导突起的生长并发挥重要作用。[著者文摘]

Expression and significance of GAP-43 gene during differentiation of hippocampus-derived neural stem cells in rats

Abstract:
Objective: To investigate the expression and significance of GAP-43 gene during the differentiation of hippocampus-derived neural stern cell ( NSC ) in rats. Methods: NSC was obtained from the hippocampus of newborn rats and identified by nestin immunocytochemistry. The expressions of GAP-43 mRNA and protein were determined by reverse tmnscriptase polymerase chain reaction and immunohistochemical method at different time points during the differentiation. Results:The expression of GAP-43 mRNA was not detected in the neurosphere,expressed at a low level at the 12th hour of differentiation,reached the peak on day 3,and then decreased on day 5. The expression of GAP-43 protein was located in the cytoplasm of NSC on day l, reached the peak and was mainly located in the prominence on day 5, and decreased gradually on day 7. Conclusion: GAP-43 plays an important role in the growth of prominence during the early period of differentiation of NSC into mature neurons.[著者文摘]
