MTT法探讨改良五黄油对人皮肤成纤维细胞增殖的抑制作用

摘 要:
目的MTT法探讨改良五黄油对人皮肤成纤维细胞增殖的抑制作用，为烧伤创面外用药物五黄油的改良目的之一——抑制瘢痕增生，寻求确切有效的实验依据。方法倒置显微镜下观察细胞形态学变化。MTT比色法检测不同终浓度（0．10、0．15、0．20、0．25、0．30g／ml）改良五黄油作用下增殖状态下成纤维细胞72h后时细胞增殖的抑制程度，主要指标，细胞形态，吸光度（A）值。结果倒置显微镜下可见空白时照组成纤维细胞大小均匀，生长密集，改良五黄油实验组和五黄油对照组细胞生长及形态明显改变，增殖缓慢，生长差，贴壁能力减弱。MTT比色法结果表示：0．10～0．30g／ml浓度药物作用人成纤维细胞，其增殖抑制程度与改良五黄油的浓度存在正相关性；与五黄油相对照，改良五黄油增殖抑制率以0．30g／ml浓度最显著（P〈0．01）。结论体外抑制人成纤维细胞增殖，改良五黄油比五黄油更有效。[著者文摘]

Influence of Modified Wuhuang Oil on The Proliferation of Fibroblasts in Vitro

Abstract:
Objective To investigate the influence of ＂ Modified Wuhuang oil＂ on the proliferation of the human fibroblasts （HDF） in vitro and the mechanism of the inhibition. Methods Cultured HDF were exposed to different concentration of ＂Modified Wuhuang oil＂solution and investigated by MTT assay after exposure to ＂Modified Wuhuang oi1＂72 hours. Using MTT assay observed the effect of ＂Modified Wuhuang oil＂on the cells growth at different times and in different concentrations（0.0-0.30g/ml）. Results ＂Modified Wuhuang oil＂ （0.10-0.30g/ml） significantly suppressed the proliferation of HDF in vitro and showed a dosedependent tendency exposure to ＂Modified Wuhuang oil＂. Conclusion ＂ Modified Wuhuang oil＂has an effective inhibition on the proliferation of HDF may supply a potential prospect in future clinical application.[著者文摘]
