黄芪诱导骨髓间充质干细胞向神经干细胞分化

摘 要:
目的:观察黄芪注射液诱导大鼠骨髓间充质干细胞(BMSCs)分化的特点以及黄芪对BMSCs活性的影响。方法:分别使用浓度为20、50、100、200μl/ml的黄芪注射液诱导BMSCs,分别在1、5、24h光镜观察细胞形态变化,免疫细胞化学方法观察巢蛋白、神经元特异性烯醇化酶(NSE)、胶质纤维酸性蛋白(GFAP)、微管相关蛋白2(MAP-2)的表达,MTT方法检测黄芪对BMSCs活性的影响。结果:诱导后细胞体积增大,胞核固缩,核质比减小,有细长突起形成,部分细胞间可见网络状连接。巢蛋白阳性细胞在100/μl/ml浓度诱导24h组染色最强,而NSE和GFAP阳性细胞在200/μl/ml浓度诱导24h组阳性细胞数量最多,染色最强,MAP-2抗体染色只在100/μl/ml浓度诱导24h组和200/μl/ml浓度诱导24h组出现少量阳性细胞。不同浓度的黄芪注射液以及作用不同时间均对BMSCs的活性产生影响。结论:在黄芪作用下,BMSCs表达神经干细胞特异标志物,随着时间的延长,进一步表达神经元特异标志物和胶质细胞特异标志物。[著者文摘]

Abstract:
Objective: To observe the differentiation of tragalus mongholicus. Methods : BMSCs were induced bone marrow mesenchymal stem cells (BMSCs) induced by asby the different concentration of astragalus mongholicus : 20μl/ ml, 50μl/ml, 100μl/ml, 200 /μl/ml. At 1, 5, 24 h after induction, the morphological changes of BMSCs were observed with optical microscope and the expressions of Nestin, NSE, GFAP and MAP-2 were identified by immunocytochemical techniques. Results: After induction, the volume of cells was increased, the nucleus of cells was shrunk, the ratio of nucleus and plasma was decreased, the long and thin dendrites were formed and some cells had reticular joint. The Nestin positive straining was strongest in the group of 100 μl/ml-24 hours, the NES and GFAP positive straining were strongest in the group of 200 μl/ml-24 hours, and the MAP-2 positive straining was only found in the groups of 100 μl/ml-24 hours and 200 μl/ml-24 hours. Conclusion: With the induction of astragale astragale, BMSCs can express the special protein of neural stem cells (Nestin). As the induction goes on, they can express the special proteins of neurons (NSE and MAP-2) and glial cells (GFAP).[著者文摘]
