Tau40蛋白的纯化及其多克隆抗体的制备

摘 要:
目的快速纯化tau蛋白,制备tau蛋白的多克隆抗体。方法依次使用差速离心、热失活、离子交换层析的方法纯化体外重组的人tau蛋白;用此纯化的蛋白免疫家兔制备多克隆抗体,用Western blot检测组织来源的样品,确定所制备抗体的特异性和效价。结果采用上述方法成功获得人类全长tau蛋白——tau 40;同时,所制备的tau蛋白多克隆抗体效价高,特异性好。结论本方法所获得的tau抗体效价高,特异性好,可以用于进一步的科学研究。[著者文摘]

Abstract:
Objective To quickly purify tao protein and produce multiclonal antibody of tau protein. Methods Human recombinant tau protein in vitro was purified by using differential centrifugation, heat inactivation and ion exchange chromatography. The rabbits were immunized with the purified tau protein to produce multiclonal antibody. The titers and specificity of the antibody were detected by using Western blot. Results The human full-length tau protein, tau40, was obtained. The multiclonal tau antibody was yielded with high titer and specificity. Conclusion The tau multiclonal antibodies obtained have high titers and specificity.[著者文摘]
