摘  要:

采用鄂报春的嫩叶片进行诱导试验,结果表明:被诱导的鄂报春叶片先形成愈伤组织再形成芽;诱导叶片分化的较理想培养基是:MS+6-BA2.0mg/L+NAA1.0mg/L,其诱导分化率可达42.11%;较理想的继代增殖培养基是:MS+6-BA1.0mg/L+NAA0.05mg/L,其有效芽分化系数为4.27,平均苗高为1.5cm;较理想的生根培养基为MS基本培养基,平均根条数为8.6条,平均根长为3.5cm,平均苗高为5.0cm,根粗壮,有利于移栽;采用培养料体积比为7:3混合的新鲜河泥和珍珠岩移栽鄂报春,成活率可达98.1%.[著者文摘]

Abstract:

A tissue culture experiment with Primula obconica was carried out and tender leaves were used as the initial explant to induce differentiation. The result revealed that the induced tender leaves of Primula obconica would form callus first and then differentiate into buds. The comparatively suitable medium for young leaf induction was MS + 6 - BA 2.0 mg/L + NAA 1.0 mg/L, on which the average inducing rate reached 42.11%. The better medium for successive culture was MS + 6 - BA 1.0 mg/L + NAA 0.05 mg/L, on which the average differentiation coefficient for effective buds reached 4.27, and the mean height of young shoots was 1.5cm. Ordinary MS medium was the best for rooting, on which the average number of roots occurred was 8.6, with the average root length as 3.5 cm, and the average shoot height as 5.0 cm. The roots induced by the ordinary medium were strong enough for transplanting. The material for Primula obconica transplanting was the combination of river mud and perlite ( proportion 7 : 3 ) , the average survival rate could reach 98.1% in the experiments.[著者文摘]
