转BtCry3A基因杨树毒蛋白表达及对桑天牛抗性的研究

摘 要:
为探索转抗虫基因(BtCry3A)741杨对鞘翅目昆虫的抗虫性,以7个转基因株系为检测对象,从分子生物学检测、毒蛋白的表达以及抗虫性检测等方面对转抗虫基因(BtCry3A)741杨株系进行了比较分析。7个转基因株系的DNA经PCR扩增显示,已转入BtCry3A基因。不同时期测定转基因株系不同部位叶片的毒蛋白含量不同,5月份测定的毒蛋白含量在当年生枝条不同部位叶片中未表现出明显的差异,至9月份封顶前,毒蛋白含量明显降低,且顶部叶片含量显著高于中下部叶片。在枝条的木质部中,毒蛋白的相对含量明显高于韧皮部。转基因株系对桑天牛幼虫的致死率很低,但对幼虫的发育具有明显的抑制作用。[著者文摘]

Expressions of BtCry 3A Gene in Transgenic Polar and its Resistance Against Apriona germari

Abstract:
To investigate the resistance against coleopterous insects of seven clones of transgenic 741 poplar, we examed them at molecular level, and analyzed the expression of BtCry3A gene. The results showed that Bt toxin protein content in different of clones and parts were different. There was no obvious regularity in examined leaves on twigs of 1 year old in May. In September, the content of toxins decreased obviously. The content in the upper leaves was higher than that of the lower and the middle, and that of xylem was higher than that of phloem. The clones had lower lethality to Apriona germari, but had obvious resistance against its larvae.[著者文摘]
