野桑蚕和不同家蚕品系幼虫体内1-脱氧野尻霉素含量测定

摘 要:
采用反相高效液相色谱(RP-HPLC)-紫外检测法测定了野桑蚕和不同品系家蚕体内的1-脱氧野尻霉素(1-DNJ)含量,并调查了家蚕5龄幼虫不同组织不同生长时期1-DNJ含量的变化。野桑蚕与家蚕、家蚕不同品系间的1-DNJ含量存在极显著差异:野桑蚕全蚕粉中1-DNJ含量最高,质量分数平均为0.4278%;家蚕以黄血蚕最低,平均0.2205%。家蚕5龄幼虫不同组织的1-DNJ含量也存在明显差异:家蚕血干粉中的1-DNJ含量最高,质量分数达0.8487%;其次是中肠、体壁和脂肪体组织,分别为0.5122%、0.4722%、0.3085%;而在丝腺中几乎检测不出1-DNJ。家蚕5龄幼虫体内1-DNJ含量随发育时期呈明显变化,5龄第4天含量最高,达0.329%,第5天以后迅速下降。[著者文摘]

Investigation of 1-DNJ Contents in the Larvae of Bombyx mori and Bombyx mandarina

Abstract:
In this paper, the 1-deoxynojirimycin(1-DNJ) contents in the 5th instar larvae at different developmental stages and tissues of Bombyx mori and Bombyx mandarina were investigated by Reverse-phase High-performance Liquid Chromatography. The results showed that there were obvious 1-DNJ changes in larvae between Bombyx mori and Bombyx mandarina, as well as among various Bombyx mori strains. The highest 1-DNJ content was found in Bombyx mandarina larvae and the average is 0. 427 B%. The domesticated silkworm with yellow blood has the lowest 1-DNJ content, and the average level is about 0. 220 5%. The 1-DNJ content of translucent silkworm larvae is extremely higher than that of the normal one, and the normal is higher than that with yellow blood. The differences of 1-DNJ content among different tissues are obvious. The highest is in the blood, the next is in the midgut, fat body, and skin, in order. The silkgland almost has no 1-DNJ. The 1-DNJ content of domesticated silkworm larvae changes significantly with developmental stages. It reached a peak of 0. 329% at the 4th day of the 5th instar and then decreased quickly.[著者文摘]
