高效液相色谱法检测人血浆中甲砜霉素及其药动学研究

摘  要:

目的:建立检测人血浆甲砜霉素的高效液相色谱法,并对其在人体内的药代动力学进行研究。方法:以ZORBAX SB—C18(4.6mm×250mm,5μm)为色谱柱;乙腈-水(30:70)为流动相,流速0.8mL·min^-1;以10,11-二氢-10-羟基-卡马西平(MHD)为内标,血浆样品经乙酸乙酯和正己烷提取后,在225am波长下进行检测。结果:甲砜霉素在0.1~10.0mg·L^-1浓度范围内线性关系良好(r=0.9999),定量下限为0.1mg·L^-1;0.25,1.0,5.0μg·mL^-1 3种浓度的日间、日内精密度RSD(n=5)均小于10%;相对回收率均在85%~115%范围内,绝对回收率大于75%。结论:本方法操作简便,结果准确;适用于血浆甲砜霉素的检测。[著者文摘]

Abstract:

Objective:To establish an HPLC method for the determination of thiamphenicol in human plasma and study of its pharmacokinetics in Chinese volunteers. Method:The analytical column was packed with ZORBAX SB -C18 (4. 6 mm × 250 mm ,5 μm) ,the mobile phase was acetonitrile -water (30: 70) ,the flow rate was 0. 8 mL ·min^-1, and detection wavelength was set at 225 nm, respectively. Plasma samples were extracted with ethyl acetate and n - hexane. Results: The linear range of thiamphenicol in human plasma was 0. 1 - 10. 0 mg · L^-1 ( r = 0. 9999). The lowest limit was 0. 1 mg · L^-1 ; Intra - day and inter - day RSD for the analysis of thiamphenicol in plasma samples at three concentration levels were both lower than 10%. Relative and absolute recoveries were 85% -115% and more than 75%, respectively. Conclusion: The HPLC method is simple, high selective and sensitive for the determination of thiamphenicol in plasma.[著者文摘]
