Ochrobactrum intermedium DN2发酵培养基优化研究烟碱降解酶

摘 要:
研究工作中筛选得到一株有较强烟碱降解能力的新型烟碱降解细菌Ochrobactrum intermediu mDN2,对其产烟碱降解酶的发酵培养基进行了优化。采用Plackett-Burman设计法,对影响Ochrobactrum intermediumDN2产烟碱降解酶的15个因子进行了筛选。结果表明,影响该茵发酵产酶的显著因子为:烟碱、酵母膏、葡萄糖、tween-80的质量浓度和培养基初始pH。在此基础上,采用响应曲面法对以上5个显著因子进行了优化。得到各因子最佳水平为:烟碱2.183g/L,葡萄糖0.823g/L,酵母膏0.844g/L,Tween-800.976g/L,初始pH6.9。在此优化条件下获得实际酶活为7943U/L,与预测值8060U/L相近,比优化前提高了52%。[著者文摘]

Media Optimization of Ochrobactrum intermedium DN2 for Nicotine-Degrading Enzyme

Abstract:
In previous study, it was found that Ochrobactrum intermedium DN2 was a kind of bacteria which can deyrad nicotine. Plackett-Burman design was used to evaluate the importance of the fifteen factors which may affect the production of nicotine-degrading enzyme of DN2. The significant factors were listed as follows: nicotine, glucose, yeast extract, Tween-80 and initial pH. Then the central composite design and response surface analysis were used to determine the optimal levels of the five significant factors. The optimized conditions were listed as follows: nicotine 2. 183g/L,glucose 0. 823 g/L yeast extract 0. 844 g/L Tween-80 0. 976 g/L,initial pH6. 9. With the optimum conditions, a highest enzyme activity was achieved to (8 010 U/L), which was close to the predicated value 7943U/L and was high 52% than that of the control.[著者文摘]
