PCR-RFLP分析用的梨基因组DNA提取

摘 要:
采用改良CTAB法,提取了23个梨品种基因组DNA。结果表明:所提取DNA产率在200μg/g鲜叶以上,大部分梨品种OD260/OD280在1.8左右,用于PCR-RFLP分析时,PCR扩增产物谱带清晰,且PCR扩增产物能被相应的限制性内切酶酶切。[著者文摘]

Abstract:
The genomic DNA of 23 pear cultivars was extracted from young leaves by an improved CTAB method. The average yield of DNA was over 200 μg/g by fresh leaves. The value of OD260/ OD280 for most pear cultivars ranged from 1.7 to 1.9. These results indicated that the purity of the extracted DNA was suitable for PCR-RFLP analysis. The PCR amplification profiles were distinct. The PCR fragments could be digested with corresponding restriction endonucleases.[著者文摘]
