中国东南近海斜带石斑鱼群体的地理分化和遗传结构研究

摘  要:

以来自3个地区55尾斜带石斑鱼(Epinephelus coioides)为研究对象,通过测定和分析长度为492bp的线粒体细胞色素b的部分基因序列,确定了10种单倍型和8个多态位点。结果表明各地理群体多态性均较低,并且单倍型在群体间分布不均,台湾群体多态性最高,海南群体次之,广东群体最低。各群体内Tajima’s D测验结果不显著(P〉0.10),符合中性突变,即群体大小维持稳定并未出现扩张和持续增长。各群体间均呈现显著的遗传分化和较低基因流水平。单倍型网络关系图也表明地区间的地理分化和单起源多分支的格局,分析形成原因可能是偶然事件中个体的长距离迁移和群体的瓶颈效应。[著者文摘]

Genetic structure and geographic subdivision of orange-spotted grouper (Epinephelus coioides ) in coastal waters of Southeast China

Abstract:

The 492 bp fragments of the mitochondrial cytochrome (cyt b ) gene of Epinephelus coioides from three geographic regions were sequenced and analyzed. It was found that there were 10 haplotypes and 8 nucleotide variable sites in 55 individuals of E. coioides. The results showed that all the population from there geographic regions exhibited low levels of nucleotide diversity and haplotype diversity, in which the population of GuangDong area were the lowest. Selective neutrality tests of the Tajima' s D were not significant( P 〉 0.10). The data suggested that there was not a population expansion of the E. coioides in these three areas, and the size of the populations was relatively stable. Analysis demonstrated a low level of gene flow and distinct genetic differentiation in different geographical regions. Minimum-spanning network analysis revealed a star-shaped pattern, which suggested that populations had rooted from only one haplotype and undergone the differentiation. The results show that Bottleneck effect and/or immigration by jump dispersal events across large geographical distances are likely the major factors in shaping such genetic structure.[著者文摘]
