摘 要:
目的研究PP333对野葛常温和低温保存的影响。方法通过常温培养法和4℃低温培养法探讨PP333对野葛离体保存的影响。结果在常温和4℃低温下,添加PP333均可提高野葛试管苗的成活率。在常温下,PP333与2mg/L6-BA、1mg/LNAA组合使用的效果比单独使用的效果要好,野葛的最佳PP333浓度分别为8mg/L(与2mg/L6-BA、1mg/LNAA组合使用)和5me/L(单独使用),180d后成活率可达80%以上;在4℃低温下,PP333使用的效果比常温保存使用的效果要好,野葛的最佳PP333浓度为5mg/L(与2mg/L6-BA、1mg/L NAA组合使用),180d后成活率可达98%。离体保存后的试管苗转入继代培养基中进行恢复培养,其生长情况与正常继代苗无显著差异。结论适宜浓度的PP333结合4℃低温可以提高试管苗的成活率,达到野葛离体保存的目的。[著者文摘]

Abstract:
Object To study the effects of PP333 on preservation in vitro of Pueraria lobata .Methods The effect of PP333 on preservation in vitro of Pueraria lobata were explored by room temperature and 4℃ low temperature culture methods. Results At room and low temperature) PP333 could improve revival rates of Pueraria lobata plantlets. At room temperature) the treatment effect of PP333 with 2 mg/L 6 - BA and 1 mg/L NAA was better than that of PP333 alone. The best PP333 concentrations of Pueraria lobata were 8 mg/L (treated by PP333 with 2 mg/L 6- BA and 1 mg/L NAA) and 5 mg/L (treated by PP33a alone) respectively, its revival rate was up to 80% after 180 days; At 4℃ low temperature, its treatment effect of PP333 was better than that of PP333 at room temperature. The best PP333 concentrations of Pueraria lobata was 5 mg/L ( treated by PP333 with 2 mg/L 6 - BA and 1 mg/L NAA), its revival rate was up to 98% after 180 days. The plantlets were cultured at room temperature that transferred to subculture medium after conserved in vitro , Pueraria lobata regenerated plantlets were as same as the normal subculture plantlets in morphology. Conclusion The suitable concentration of PP333 with 4℃ low temperature can improve evidently revival rates of Pueraria lobata plantlets.[著者文摘]
