产蛋白酶生防细菌的筛选及其对病原真菌的拮抗作用

摘 要:
从小麦根际土壤中分离到一株高产蛋白酶并对多种植物病原真菌具有拮抗作用的生防细菌T2。通过形态特征、生理生化及16S rDNA同源性序列分析,初步鉴定该菌为枯草芽孢杆菌(Bacillus subtilis)。其发酵液经硫酸铵沉淀后的粗酶液可明显抑制棉花枯萎病菌菌丝生长及孢子萌发,在显微镜下观察到棉花枯萎病菌孢子萌发的芽管短、孢子膨大变形呈泡囊状,菌丝呈串珠状膨大、细胞破裂、细胞质外渗,表明该细菌菌株产生的胞外蛋白酶可能与病菌菌丝生长受抑制密切相关。[著者文摘]

Abstract:
Bacterial strain T2 was a potential biocontrol agent against a broad spectrum of plant pathogenic fungi with the ability to produce extracellular protease. According to the characteristics of morphology, physiology and biochemistrytests and the comparison of 16S rDNA sequence , the strain T2 was similar to Bacillus subtilis. The crude protease obtained by precipitating the cell-free culture of T2 with 80% saturated (NH4)2SO4 could inhibit the mycelial growth and the germination of conidia, including morphological abnormalities such as swelling, distortion, it may be inferred that the extracellular protease produced by Bacillus subtilis is closely related to the inhibition of hypha growth.[著者文摘]
