摘 要:
本文旨在探讨牛膝多糖(ABPS)对断奶仔猪抗菌肽Protegrin-1(PG-1)mRNA表达的影响。试验采用单因子设计,按照日粮处理因素不同,设对照组、抗生素组、0.05%、0.10%和0.15%牛膝多糖组,共5个处理。选用28日龄社长大仔猪160头,随机分人5个处理,每个处理4个重复,每个重复8头猪(公母各半)。采用半定量RT-PCR法,以18SrRNA为内标,检测PG-1mRNA相对表达量。结果表明:日粮中添加牛膝多糖均不同程度地促进断奶仔猪骨髓抗菌肽PG-1mRNA的表达。与对照组相比,0.05%、0.10%和0.15%牛膝多糖组分别使断奶仔猪PG-1mRNA的相对表达量增加14.9%(P〉0.05)、36.4%(P〈0.05)和7.3%(P〈0.05);与抗生素组相比,其PG-1mRNA的相对表达量分别增加7.8%(P〉0.05)、27.9%(P〉0.05)和19.4%(P〉0.05)。即牛膝多糖能显著促进断奶仔猪骨髓抗菌肽PG.1mRNA表达,且日粮中以0.10%添加量为适宜。[著者文摘]

The Effect of Achyranthes Bidentata Expression in Polysaccharides on Protegrin-lmRNA Weaned Piglets

Abstract:
To investigate the effect of A.chyranthes bidentata polysaccharides(ABPS) on expression of antibacterial peptide Protegrin-1 (PG-1) mRNA in weaned piglets,according to the different dietary factors, five test groups were used, including control group, antibiotics group, 0,05%, 0.10% and 0, 15% ABPS groups. One hundred and sixty weaned piglets (Duroc x (Landrace x Yorkshine)) with age of 28 days were randomly allotted to five test groups with four replicates in each group and eight weaned piglets in each repeat. An optimized semi-quantitative RT-PCR was constructed to detect the relative ex- pression levels of PG-1 mRNA by using 18S rRNA as inner control. The results showed that PG-1 mRNA expression could bc enhanced to a different extent with ABPS in weaned piglets, Comparcd with control group, 0.05%, 0.10% and 0, 15% AB- PS enhanced the PG-1 mRNA expression by 14.9 % ( P〉0.05), 36.4% (P〈0.05) and 27.3 % ( P〈0.05) respectively; Compared with antibiotics group, 0. 05%, 0. 10% and 0. 15% ABPS increased the PG-1 mRNA expression by 7.8% (P〉 0.05 ), 27.9% (P〉0.05) and 19.4% (P〉0.05) respectively. Above results suggested that, ABPS remarkably increased rel- ative expression of PG-1 mRNA in femur marrow of wcaned piglets, and the appropriate ABPS addition was 0.10%.[著者文摘]
